چکیده

The intraerythrocytic malaria parasite constructs an intracellular haem crystal, called haemozoin, within an acidic digestive vacuole where haemoglobin is degraded. Haem crystallization is the target of the widely used antimalarial quinoline drugs. The intracellular mechanism of molecular initiation of haem crystallization, whether by proteins, polar membrane lipids or by neutral lipids, has not been fully substantiated. In the present study, we show neutral lipid predominant nanospheres, which envelop haemozoin inside Plasmodium falciparum digestive vacuoles. Subcellular fractionation of parasite-derived haemozoin through a dense 1.7 M sucrose cushion identifies monoacylglycerol and diacylglycerol neutral lipids as well as some polar lipids in close association with the purified haemozoin. Global MS lipidomics detects monopalmitic glycerol and monostearic glycerol, but not mono-oleic glycerol, closely associated with haemozoin. The complex neutral lipid mixture rapidly initiates haem crystallization, with reversible pH-dependent quinoline inhibition associated with quinoline entry into the neutral lipid microenvironment. Neutral lipid nanospheres both enable haem crystallization in the presence of high globin concentrations and protect haem from H2O2 degradation. Conceptually, the present study shifts the intracellular microenvironment of haem crystallization and quinoline inhibition from a polar aqueous location to a non-polar neutral lipid nanosphere able to exclude water for efficient haem crystallization.

Genotyping of C and F Regions of Plasmodium Falciparum EBA-175 in South-East of Iran

Abstract Background and Objective: The Plasmodium falciparum EBA-175, via Sialic acid dependent glycophorin A, binds to red blood cells and thus plays a critical role in cell invasion. Some part of second allele in its gene encoding in FCR-3 (Section F) and CAMP (Section C) can be found. This study aimed to determine the prevalence of Plasmodium falciparum EBA-175KD alleles in southeastern I...

Clinical Pharmacology of the Antimalarial Chloroquine in Children and Their Mothers

Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are the parasites that infect humans. Plasmodium falciparum and Plasmodium vivax cause most of the malarial infections worldwide. Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are susceptible to chloroquine. Chloroquine was the world's most widely used antim...

Biliverdin targets enolase and eukaryotic initiation factor 2 (eIF2α) to reduce the growth of intraerythrocytic development of the malaria parasite Plasmodium falciparum

In mammals, haem degradation to biliverdin (BV) through the action of haem oxygenase (HO) is a critical step in haem metabolism. The malaria parasite converts haem into the chemically inert haemozoin to avoid toxicity. We discovered that the knock-out of HO in P. berghei is lethal; therefore, we investigated the function of biliverdin (BV) and haem in the parasite. Addition of external BV and h...

Genetic Diversity Block 2 of Surface Protein-1 in Plasmodium Falciparum Merozoite by Nested-PCR Method in Southeastern Iran

Abstract       Background and Objectives: Plasmodium falciparum merozoite surface protein-1 (PfMSP-1) is a promising vaccine against malaria during its blood stages which play an important role in immunity to this disease. Polymorphic nature of this gene is a major obstacle in making an effective vaccine against malaria. In this study, the genetic diversity of Plasmodi...

C1 Inhibitor, C3 Activator, IgG, IgA, and IgM Titers in Nigerian Sickle Cell Disease Patients with Plasmodium falciparum

Background: Sickle cell disease (HbSS) is a major health problem in Nigeria and ma-laria has been implicated as a leading cause of morbidity/mortality in sickle cell disease patients. Few reasons were put forward to explain the observed morbidity/mortality of HbSS subjects due to Plasmodium falciparum (P. falciparum) malaria. Objectives: To determine the level of immunoglobulin classes (IgM, Ig...
